When John Perrine and his twin brother John were born on February 14, 1744, in Perth Amboy, New Jersey, their father, Matthew, was 34, and their mother, Isabella, was 26. He married Elizabeth Forman and they had one daughter together. He then married Eleanor Wyckoff and they had four children together.

In the American Revolution he served as a Sergeant under Capt Jacob Ten Eyck’s Somerset Company, in the New Jersey 1st Battalion

He died on June 14, 1809, in East Windsor, New Jersey, at the age of 65, and was buried in Cranbury, New Jersey.
