Joseph Doane was born in 1720 in Eastham, Barnstable County, Massachusetts, the son of Elisha and Hannah (Cole) Doane.

On 30 August 1775, he was commissioned to serve as the Colonel of the 2nd Barnstable County Regiment. He was serving in that capacity through at least 1777.

He married Dorcas Eldridge on 12 April 1739 in Eastham, Barnstable, Massachusetts. His estate probate lists a widow named Hannah, which suggests his first wife had died and he remarried. His known children are thought to have included:
